Detection principle

Sialic acid forms a purplish red complex with methyl resorcinol in the presence of oxidant. And the absorbance conforms to Lambert-Beer's law. The content of sialic acid can be calculated by measuring the OD value at 560 nm. When detect tissue and cell samples, the protein concentration of the sample needs to be detected in addition (E-BC-K165, E-BC-K168, E-BC-K318 are recommended).

Dilution of sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.022-7 mmol/L).

Asked: Dear Sir/Madam, My question is regarding the applicability of this kit to detect the artificial sialic acid for example: 9-azido-sialic acid. Can you please first describe the exact mechanism or chemical reaction responsible for the color production, since its not explained anywhere in the ELabscience website. Next, can you please guide whether it would be applicable to determine sialic acid on the proteins, when decorated with 9-Azido-sialic acid.

Answered: The color development principle of E-BC-K068-S kit is that Sialic acid forms a purplish red complex with methyl resorcinol in the presence of oxidant. And the absorbance conforms to Lambert-Beer's law. The content of sialic acid can be calculated by measuring the OD value at 560 nm. The protein decorated with 9-Azido-sialic acid can react with methyl resorcinol. However, the laboratory did not validate this sample, and in theory, as long as the sialic acid content of the sample exceeds the sensitivity (0.022 mmol/L) of the kit, it can be detected.
