Detection principle

ALT catalyze the amino conversion reaction between alanine and α-ketoglutaric acid to produce pyruvic acid and glutamic acid at pH 7.4 and 37℃. Then phenylhydrazine was added to form phenylhydrazone with pyruvic acid. Phenylhydrazone is reddish brown under alkaline conditions. ALT activity can be calculated by measuring the OD values at 510 nm.

Images

No significant change of ALT activity was observed, the LCFG nanoparticles did not include hepatotoxicity.

X Zhang et al investigate the usage of H2O2 and O2 for cooperative chemodynamic/starvation cancer therapy. Alanine aminotransferase (ALT) avtivity in mouse blood was determined using ALT activity assay kit (E-BC-K235-M).

Dilution of sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.75-72.3 IU/L).

The recommended dilution factor for different samples is as follows (for reference only):

Asked: How to collect cell culture supernatant and perform subsequent detection experiments, and what is the difference between supernatant and direct use of cell lysis buffer?

Reply

A

adminSubmitted [ Aug 04 2023 ]

Answered: 1. The preparation of cell culture supernatant: Suspension cells: Centrifuge the cell suspension 1000g for 10 minutes, and take the supernatant on ice for detection. Adherent cells: directly absorb the supernatant, centrifuge 1000g for 10 minutes, and place on ice for detection. 2. The cell supernatant reflects the enzyme activity on the outside of the cell, while the cells treated with the cell lysis buffer reflect the enzyme activity inside the cell.

Asked: Can you please help me understand exactly what a carmen unit is and why we are using it instead of the conventional method of plotting a standard curve of concentration of standard vs the absorbance. How is the carmen unit relevant to this exact calculation I would also like to know how you arrived at the numbers 0, 28, 57, 97, 150, 200 for the standard Carmen values.

Answered: 1. This indicator is special, and the method used is the Reitman-Frankel method. The standard curve of the Reitman-Frankel method is determined by comparing the colorimetric method experimental result with the result of karmen spectrophotometry (rate method), and the report result of Karmen units will be more accurate. 2. The Karmen's units corresponding to the standard are fixed and converted according to the concentration of pyruvate.
