Application This ELISA kit applies to the in vitro quantitative determination of Human Zonulin concentrations in serum, plasma and other biological fluids.

Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human Zonulin. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human Zonulin and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human Zonulin, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human Zonulin. You can calculate the concentration of Human Zonulin in the samples by comparing the OD of the samples to the standard curve.

The recovery of Human Zonulin spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Assay Procedures

1. Add 100μL standard or sample to the wells. Incubate for 90 min at 37°C

Answered: You can dilute the sample using the Reference Standard & Sample Diluent in the kit. When the sample is not diluted, it is not necessary to use standard product & sample diluent for sample loading. Sample dilution of 5 times requires at least 20ul sample +80ul standard & sample diluent. The loading volume of the sample is 100ul.

Answered: For E-EL-H5560 kit, the reference value of normal human serum samples is 40-115ng/ml, according to our internal verification result. However, considering about the individual differences, we recommend that you set up your own normal sample group as control in order to determine whether the zonulin levels are elevated.

Asked: In your previous answers to other questions, you said, \"the immunogen sequences of the capture antibody and detection antibody were human haptoglobin 19-406aa (#P00738).\" So that would mean that haptoglobin could produce a false positive or falsely elevated \"zonulin\" result in your test. How do you know you are detecting zonulin/pre-HB2 and not haptoglobin? You also said, \"The antigen is full-length recombinant pre-HP2. The expression host of the antigen is HEK 293 cell.\" Which antigen are you referring to in that second statement, since you already stated that the antigen which the capture antibody is specific for is haptoglobin rather than zonulin/pre-HP2? If the capture and detection antibodies react with both haptoglobin and pre-HP2, how is your assay specific for only pre-HP2? Thank you.

Answered: We applied anti-ZOT antibodies in this kit, ZOT is an enterotoxin elaborated by Vibrio cholera, a zonulin analogue. It has been reported that this antibody specifically recognizes the human zonulin (pre-HP2) instead of the mature Haptoglobin. In addition, we verified that the kit does not have significant cross-reactivity with mature HP protein.

Answered: The zonulin protein is the inactive precursor for HP2 and it is an uncleaved single-chain form. For our kit, it can detect the Precursor of haptoglobin 2, the immunogen sequences of the capture antibody and detection antibody were human haptoglobin 19-406aa (#P00738). Answered: Thanks for your message. E-EL-H5560 Human Zonulin ELISA Kit Yes, the kit can recognize zonulin, but cannot recognize complement C3. We also found the document you mentioned in the attachment, these words don't mean one kit can recognize two proteins (zonulin, complement C3), which means the CUSABIO kit can detect serum zonulin levels, and our understanding literature says is human complement C3c, HP1-1, HP2-2 and recombinant zonulin (zon) standards were separated by SDS-PAGE and stained with Coomassie gel stain . Answered: Dear Hayashi san,thanks for your message. This kit is specifically designed to detect Zonulin. The kit can be used to detect natural Zonulin. In theory, the kit also can be used for recombinant Zonulin. However, due to the influence of the immunogen sequence and the expression host of the recombinant protein, some recombinant Zonulin may not match the antibodies in the kit. Therefore, a pre-test is recommended. The antigen is full-length recombinant pre-HP2. The expression host of the antigen is HEK 293 cell. Best Regards, Y
