Fast Coomassie Blue Staining Solution (10×)

Introduction

Elabscience^® Fast Coomassie Blue Staining Solution uses coomassie blue G250 as the dye. It could stain SDS-PAGE or Native-PAGE gels rapidly. This solution is non-toxic, no pungent smelling, with integrates fast, high precision, easy to destain and stability.

Fast: Wash the gel for 5 min and stained for 10 min, and the electrophoresis bands could be observed.
High precision: 200 ng bands can be detected after 10 min of staining, refer to Figure 1a. However it is difficult to detect by conventional staining, refer to Figure 1b.

Low background: The bands can be observed without destaining, and the low background images can be obtained by destaining with DI water, extend destaining time could be better. Refer to Figure 2.

Stability: Storage of the solution at 37℃ for 2 weeks has no significant difference in dyeing effect, refer to Figure 3.

Instructions

Fast Coomassie Blue Staining Solution(10×) is a 10 × concentrated solution. Dilute with DI water to 1 ×Fast Coomassie Blue Staining Solution before use.

For example: Take 10 mL Fast Coomassie Blue Staining Solution(10×), dilute with DI water to 100 mL.

Experimental Procedure

1. Wash the gel

Put the gel in a container, and wash the gel with DI water. Then add about 50 mL of DI water, and heat for 3 min with a microwave or add boiled DI water and shake for 5 min, discard the DI water. It can be repeated once.

Tips: For low-concentration gels, it’s recommended to add boiled DI water instead of microwave heating. Native -PAGE can be dyed directly without this step.

2. Staining

Discard the liquid, add the diluted 1× Fast Coomassie Blue Staining Solution to make the gel immersed, heat for 30 sec with a microwave and shake at room temperature for 10 min. Discard the staining solution, and add DI water to wash the residual staining solution. Then then photograph.

Tips: 10 min is a conventional staining time. The actual staining time can be adjusted according to the result

3. Destain (Optional for lower background )

Add 50~100 mL of DI water and heat with microwave or add boiled DI water and shake to decolorize. Discard the liquid about 5 min later. It can be repeated once to get lower background.

Storage

Store at RT for 6 months in dark or store at 2~8℃ for 12 months.

Cautions

1.   This solution is acidic and corrosive slightly. For your safety and health, please wear the lab coat and disposable gloves before the experiments.

2.   It is better to use DI water, but the pure water also can be used instead of DI water.

3.   Avoid breaking the gel due when heat with the microwave
