X Zhao et al investigate the function of quercetin for reducing ethanol-induced hepatocyte pyroptosis. Total antioxidants status (TAS) of human L02 cell was determined using TAS colorimetric assay kit (E-BC-K801-M).
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.23-2 mmol Trolox Equiv. /L).
The recommended dilution factor for different samples is as follows (for reference only):

Asked: How stable are the reagents after opening the kit? How many times can the reagents be used (freeze-thaw)? Can I re-use the microplate if few wells are going to be used at a time? Thank you!

Answered: 1. After opening the kit, ensure that the reagent is sealed well and can be stored at -20℃ for 12 months. 2. Repeated freezing and thawing may have a certain impact on the properties of the reagent. If multiple tests are required, it is recommended to aliquoted storage at -20° C. 3. The microplate can not be reused. If the microplate is not enough, the microplate of this kit can be replaced by ordinary 96-well transparent plate.

Answered: E-BC-K801-M E-BC-K802-M can be used to detect serum and tissue samples, but we have only verified some routine serum and tissue samples, such as human serum, rat serum, bovine serum, and rat liver and brain tissue. In theory, TAS and TOS in fish serum tissue and muscle tissue are higher than the sensitivity of the kit can be detected.

Asked: I\'m having so much difficulty calculating TAS values (and also TOS, I bought the two kits). I\'ve already tried everything, and the values result in something very out of my understanding capacity. I would like to know if I must consider the sample\'s OD values or the mmol/L concentration. For example, when my standard curve is on 0 concentration I have a 0,049 OD concentration. Since this value is too when compared to the sample\'s OD, my results always give negative. How to proceed? Could you please assist in this or have any tutorial video that could explain more easily how to obtain these values? Thank you in advance.
