HE Staining Kit is easy to operate, add the staining solution on the sample to stain the cells. This staining solution is improved without toxic reagents such as mercury or methanol. Hematoxylin is an alkaline dye, which mainly makes the chromatin and ribosome in the nucleus purple blue, eosin is an acid dye, which mainly makes the components in the cytoplasm and extracellular matrix pink or red.
HE staining is the most basic and widely used technique in the teaching and research of histology, embryology and pathology.

Experimental Procedure
1. For Paraffin slice, conventional dewaxing to water. For frozen slice, please stain directly without dewaxing.
2. Drop Hematoxylin Staining Solution (E-IC-R117A) to cover the samples and stain for 2~5 min (the staining time can be adjusted according to the staining results and requirements).
3. Wash the slices with water.
4. Immerse the slices in the differentiation solutions for 2~30 s(the time can be adjusted according to the dyeing results and requirements), and wash the slices with water.
5. Immerse the slices in 95% alcohol for 30 s, add Eosin Staining Solution (E-IR-R117B) to cover the samples and stain for 20 s~3 min (the time can be adjusted according to the staining results and requirements).
6. Wash the slices with water.
7. Dehydration, transparency and sealing.
Storage
Store at room temperature for 12 months. Hematoxylin Staining Solution (E-IR-R117A) should be stored in dark.
Cautions
1. 4% Paraformaldehyde (It is recommended to use Elabscience® E-IR-R113), 70% ethanol and 95% ethanol, differentiation solutions (Ethanol Solution with 1% HCl) are needed in this experiment.
2. If dehydration, transparency and sealing treatment are required, xylene, Neutral Balsam (It is recommended to use Elabscience® E-IR-R118) or other sealing agent are needed in the experiment.
3. If the sample is paraffin slice, 90% ethanol, anhydrous ethanol and xylene are need in the experiment.
4. For your safety and health, please wear the lab coat and disposable gloves before the experiments.
